The 2021 season will be the San Francisco 49ers' upcoming 72nd season in the National Football League, their 76th overall and their fifth under the head coach/general manager tandem of Kyle Shanahan and John Lynch.

Notes
- Exact numbers of the selections from rounds 3–7 will be determined when compensatory selections are awarded at the NFL's annual spring owners' meetings.
- The 49ers traded their 2022 sixth-round selection to the New York Jets for defensive end Jordan Willis and 2021 seventh-round selection.[5]
- The 49ers traded Kwon Alexander to the New Orleans Saints in exchange for a fifth-round conditional pick in the 2021 NFL Draft and LB Kiko Alonso.[6]
- The 49ers traded their 2021 third-round and 2020 fifth-round selections to Washington in exchange for offensive tackle Trent Williams.[7]
- The 49ers received third-round selections in 2021, 2022, and 2023, as compensation when defensive coordinator Robert Saleh was hired by the New York Jets as head coach,[8] and vice president of player personnel Martin Mayhew was hired by the Washington Football Team as general manager.[9][10]

2021 opponents
Listed below are the 49ers' opponents for 2021. The length of the season is contingent on whether the league and the National Football League Players Association agree to expand the regular season from 16 to 17 games. If this happens, a 17th game would match the 49ers against an interconference opponent from one of the three divisions from the AFC that the 49ers are not scheduled to play in 2021, which will be based on division placement from 2020.[11]
